<div> 北京大学人民医院妇科肿瘤中心主任钱和年教授带领的课题组开展卵巢癌抗独特型抗体系统研究，目标“锁定”在阻断机体免疫逃避环节上。依据人体免疫T淋巴细胞活化的双信号学说，他们首先对6种卵巢癌细胞系及卵巢癌组织细胞进行检测，发现全部缺乏组织相容抗原II(MHC-II)及B7-1的表达，而这两种物质正是活化T细胞不可缺少的。接着，他们又采用基因工程方法成功构建了双顺反子人B7-1及干扰素-γ共表达逆转录病毒载体，即干扰素-γ可诱导MHC-II类分子，用它转导卵巢癌细胞，检测出B7-1呈高表达，干扰素-γ也呈现高水平分泌。对这种基因转导的疫苗进行体外实验，观察到它能有效地刺激T淋巴细胞活化，从而对卵巢癌细胞产生明显的杀伤力。<br />
经反复对照研究，证实了这种由双顺反子B7-1和干扰素γ共表达载体修饰的卵巢癌疫苗的作用，明显高于单独B7-1或干扰素-γ修饰的疫苗，而后两者的作用又高于对照的空载体，表明疫苗对癌细胞的杀伤作用是具有特异性的。为了得到体内验证资料，钱和年教授等人又成功建立了免疫双缺陷小鼠人卵巢癌模型，然后用人的淋巴细胞对小鼠进行免疫重建。通过这种模型实验观察，发现双修饰疫苗使小鼠致瘤性下降，发生卵巢癌的几率明显低于其它对照组，进一步证实了双基因修饰的卵巢癌细胞疫苗可增强体内的抗肿瘤免疫。从而为卵巢癌免疫基因治疗奠定了良好基础。 </div>
